We are using multiple myeloma as a model to understand the actions of glucocorticoids (GC) and the glucocorticoid receptor (GR). Development of resistance to GCs remains a critical clinical problem which involves the loss of GR expression. We are exploring post-transcriptional mechanisms to understand the loss of receptor expression. In addition we are investigating new drugs for the treatment of myeloma. We have discovered novel nucleoside analogs that are effective in both GC-sensitive and -resistant cells. We are working on bringing these compounds to clinic.
